New techniques in antivenom production and active immunization against snake venoms.

چکیده
Standard methods of commercial antivenom production are costly both in terms of animals, their maintenance, the use of large amounts of expensive venoms and man-hours. The basic technology, which involves the hyperimmunization of horses, bleeding and subsequent purification of the plasma immunoglobulin G (IgG) using salt precipitation and pepsin digestion, has not advanced significantly for many years. In most cases monospecific (animal immunized against a single venom) or polyspecilic (animal immunized against a mixture of different venoms) antivenoms are used in developing countries where snake bite is a serious problem. Funds for purchase of such drugs are of necessity limited; this can therefore result in relatively low profit margins with associated frequent inadequate quality control and substandard products.

Cross Neutralization of Afro-Asian Cobra and Asian Krait Venoms by a Thai Polyvalent Snake Antivenom (Neuro Polyvalent Snake Antivenom)
BACKGROUND Snake envenomation is a serious public health threat in the rural areas of Asian and African countries. To date, the only proven treatment for snake envenomation is antivenom therapy. Cross-neutralization of heterologous venoms by antivenom raised against venoms of closely related species has been reported. The present study examined the cross neutralizing potential of a newly develo...
متن کاملDetermination of in vivo toxicity and in vitro cytotoxicity of venom from the Cypriot blunt-nosed viper Macrovipera lebetina lebetina and antivenom production
The venomous Levantine viper, Macrovipera lebetina lebetina is endemic to Cyprus. The objective of this study was to investigate in vitro cytotoxicity, in vivo lethality, and antivenom production followed by a re-immunization schedule in mice against Macrovipera lebetina lebetina venom. The LD50 value was estimated as 7.58mg/kg within 24 hours by different venom doses administrated intraperiton...
متن کاملBioinformatics and Multiepitope DNA Immunization to Design Rational Snake Antivenom
BACKGROUND Snake venom is a potentially lethal and complex mixture of hundreds of functionally diverse proteins that are difficult to purify and hence difficult to characterize. These difficulties have inhibited the development of toxin-targeted therapy, and conventional antivenom is still generated from the sera of horses or sheep immunized with whole venom. Although life-saving, antivenoms co...
متن کاملDiversity of Micrurus Snake Species Related to Their Venom Toxic Effects and the Prospective of Antivenom Neutralization
BACKGROUND Micrurus snake bites can cause death by muscle paralysis and respiratory arrest, few hours after envenomation. The specific treatment for coral snake envenomation is the intravenous application of heterologous antivenom and, in Brazil, it is produced by horse immunization with a mixture of M. corallinus and M. frontalis venoms, snakes that inhabit the South and Southeastern regions o...
متن کاملNeutralization of the Principal Toxins from the Venoms of Thai Naja kaouthia and Malaysian Hydrophis schistosus: Insights into Toxin-Specific Neutralization by Two Different Antivenoms
Antivenom neutralization against cobra venoms is generally low in potency, presumably due to poor toxin-specific immunoreactivity. This study aimed to investigate the effectiveness of two elapid antivenoms to neutralize the principal toxins purified from the venoms of the Thai monocled cobra (Naja kaouthia, Nk-T) and the Malaysian beaked sea snake (Hydrophis schistosus, Hs-M). In mice, N. kaout...
